- Background: Forkhead box protein 01A (FOXO1a) belongs to a large family of Forkhead proteins that possesses a winged-helix DNA-binding domain called Forkhead box (FOX). FOXO1 is a transcription factor that integrates cell signals to mediate metabolism, the cell cycle, DNA repair, and response to cellular stress. In response to insulin, FOXO1 is phosphorylated by Akt and excluded from the nucleus and subsequently degraded. Over-expressed cytoplasmic FOXO1 interacts with TSC2 to cause its degradation and subsequent activation of the mTOR pathway, while nuclear localization inhibits the mTOR pathway.
